Un consorcio internacional de más de 68 científicos planea crear un “zoo del genoma”, una colección de secuencias de ADN de 10.000 especies de vertebrados, casi uno para cada género de vertebrados. Es el llamado Proyecto Genoma 10K que constituirá el estudio más exhaustivo de la evolución animal jamás intentado.  

El Proyecto Genoma 10K recogerá muestras de miles de animales de zoológicos, museos y colecciones universitarias de todo el mundo y, después, secuenciará el genoma de cada especie para desvelar su herencia genética completa. El grupo compuesto por 68 científicos y denominado Comunidad de Científicos del Genoma 10K (G10KCOS) inició el proyecto en abril de 2009 en una reunión en la Universidad de California (UC), Santa Cruz (EEUU).

“Por primera vez, tenemos la oportunidad de ver realmente la evolución en acción, justo en el cambio de los genomas completos”, afirma David Haussler, uno de los autores que concibió el proyecto, profesor de ingeniería biomolecular de la UC en Santa Cruz e investigador del Instituto Médico Howard Hughes. “Esto es posible gracias a que la tecnología para secuenciar ADN es mil veces más potente ahora que hace una década y está preparada para hacerse aún más potente muy pronto”, añade el científico.

Según Stephen J. O’Brien, director del Laboratorio de Diversidad Genómica del Instituto Nacional del Cáncer, el coste de la secuenciación del genoma ha caído durante la última década, y ha hecho posible la secuenciación de 10.000 genomas.

“El coste original de la secuenciación de un genoma humano por un gran consorcio internacional era de más de mil millones de dólares. Con la última tecnología de secuenciación, ahora cuesta entre 50.000 y 100.000 dólares por genoma. El precio sólo tiene que bajar un poco más para hacer posible la secuenciación de 10.000 genomas”, explica O’Brien.

Debido a que la evolución de las especies actuales implica viejos cambios genéticos que todavía se conservan en su ADN, el Proyecto Genoma 10K ayudaría a encontrar las respuestas a las preguntas sobre la historia de la evolución. Según los científicos, tener a mano los genomas completos permitirá estudios detallados de los cambios evolutivos de cada base a través del genoma.

“Las diferencias en el ADN que forma el genoma de los animales que encontramos hoy en día tienen la llave de los grandes eventos biológicos del pasado, tales como el desarrollo del corazón de cuatro cámaras y la magnífica arquitectura de alas, aletas y patas, cada una adaptada para su propósito especial”, manifiesta Haussler.

Entre los autores también se encuentra el genetista pionero Sydney Brenner, Premio Nobel y miembro distinguido del Instituto Salk: “El mayor desafío intelectual al que se enfrenta la biología en este siglo será la reconstrucción de nuestro pasado biológico para entender cómo evolucionan organismos tan complejos como los nuestros”, sostiene Brenner. “Los genomas contienen información del pasado – son fósiles moleculares – y, por tanto, conseguir las secuencias de los vertebrados será una fuente esencial de información valiosa”, anota el investigador.

Primera acción, crear la base de datos

En la reunión de la UC celebrada en Santa Cruz en abril 2009, unos 55 científicos en representación de zoos, museos, centros de investigación y universidades de todo el mundo determinaron los desafíos de la logística involucrada en la realización de este ambicioso proyecto.

“Se trata de científicos que han dedicado sus vidas a la biología, la evolución y la conservación de animales y ahora ven la oportunidad de realizar un estudio en profundidad”, apunta Haussler. “Salimos de aquella reunión con un plan para seguir avanzando y con una extraordinaria base de datos online con muestras de más de 16.000 especies diferentes de vertebrados recopiladas de entre más de 50 instituciones”, manifiesta el investigador.

El grupo de científicos se ha ampliado para extender la colección, y se ha creado la base de datos. El borrador de la propuesta se publica ahora en Journal of Heredity. El proyecto analizará la base de datos de genomas para desvelar los cambios evolutivos del registro, y comentará con descubrimientos experimentales relativos sitios específicos de cambio. “El análisis de estos datos supondrá un reto mucho mayor que los que hasta ahora se habían intentado en genómica comparada”, continúa Haussler.

O’Brien explica que el equipo espera que este proyecto integre la inferencia del genoma en casi todos los aspectos de la investigación biológica de los vertebrados. “La ciencia biológica trata de entender cómo funcionan las especies, así que tener disponibles los genomas de 10.000 especies dará un nuevo sentido a la forma de entender la biología”, asegura el científico.

“El genoma es como un sexto sentido que se añade a lo que podemos ver, oler, saborear, escuchar y sentir. Si disponemos de esta información para aquellas especies que por lo general no se estudian, será una flecha certera en la aljaba de los estudiantes de biología”, añade O’Brien.

Los científicos han identificado muestras que abarcan una amplia gama de la diversidad evolutiva. “Las especies incluyen mamíferos vivos, aves, reptiles, anfibios y peces, muchos de los cuales están amenazados o en peligro de extinción, así como algunas especies que se extinguieron recientemente”, asevera O’Brien que añade que los científicos están capturando lo que la evolución dejó antes de que comenzara el impacto de la población humana sobre las especies.

Entender la base genética de los vertebrados

Los participantes esperan que el Proyecto Genoma 10K siente las bases para entender la base genética de los últimos y rápidos cambios adaptativos en las especies de vertebrados y las especies estrechamente relacionadas. Los resultados pueden mejorar los esfuerzos de conservación pues los científicos podrán predecir la respuesta de las especies al cambio climático, la contaminación, las enfermedades emergentes y los competidores invasores.

“Disminuye el riesgo de extinción para aquellas especies de las que tenemos una secuencia del genoma porque esto permite estudios que pueden proporcionar información importante relativa a la conservación”, afirma Oliver A. Ryder, otro de los autores y director de genética del Instituto de Conservación e Investigación de especies en peligro del Zoo de San Diego y profesor adjunto de biología de la UC en San Diego.

Las muestras de cada especie incluyen machos y hembras, y reflejan la diversidad geográfica o la diversidad en poblaciones localizadas. La recogida incluirá más de 1.000 muestras congeladas de células fibroblásticas procedentes de 602 especies diferentes de vertebrados. Según Ryder, estas muestras de células, conservadas por el Zoo de San Diego, el Instituto Nacional del Cáncer y los repositorios de células del mundo, son un recurso valioso para los estudios de genética.

Para O’Brien, ahora empieza la parte del proyecto que exige más esfuerzos. “Hacer que esta promesa se convierta en realidad recogiendo muestras, caracterizándolas, realizando un control de calidad y entregándolas en centros de secuenciación que puedan lograr dicho objetivo supone el primer desafío”, concluye el científico. It’s interesting that after only four months after my blog opened, it has reached 10,000 hits!

Thanks you guys for visiting and especially for commenting on my site several times.

There are specific people who continously visit my blog and it’s a list that right now I am too lazy to write but thanks, you guys know who you are anyways! First, it was 1,000 in a month and now 10,000! I’m surprised and happy, I hope more people continue to read my blog more.

In fact, I’m going to take it to another level – not the layout, but I do need a change of header which I really need someone to do for me if you have the time. Also, I’m definitely not going to hold grudges with my writing now, that means comparision if I see it, opinions I never said before and even people who are find hot, I could complain about.

It has been very interesting the past several days in the stock market.  The Dow pushed over 10,000 last week and sits at nearly 10,100 today.  The weak dollar can share part of the claim reaching this important mark, but another, more interesting part are the reports by some of the country’s largest banks of very solid earnings for the past quarter.

JPMorgan reported a profit between July and September of $3.59 billion.  Goldman Sachs earned $3.19 billion during the same time period, reporting the most it has ever made in three months, with each of the bank’s employees earning an average of $700,000 EACH.  Citigroup has reported a profit of $101 million; we’ve gotten so used to the “B” word, millions seem like chump change.

I am happy to know that some companies out there are actually making a profit and helping to create some positive news out there in the market place.  However, it strikes me as completely ironic that most of these institutions are the same organizations which 1) Helped create the economic mess in which we find ourselves; 2) Were bailed out by the taxpayers, meaning the 51% and mainly the top 5% of our population which pay the majority of taxes; and 3)Have stopped the flow of capital into the market, cutting off businesses from their credit and capital lifelines, all the while paying their people unbelievable amounts of money.

Don’t get me wrong.  I am all about capitalism, free enterprise and  entrepreneurship.  Those who make it on their own and with their own drive and tenacity deserve every dollar and every success they can collect.  What I am absolutely opposed to is the organization which has an open check and safety net from the government, hoards its money by investing in T-bills, stops the flow of capital and the monetary cycle to the market, all the while rewarding itself in the process.

No matter what positive signs the media is portraying, the every day reality is that business is hurting.  My circle of friends consists of nearly all business owners; not one of them I know is looking to hire anyone soon, and may still have more downsizing to go.  Nearly all of them need capital, and nearly all of them cannot find it, leaving them with dwindling options to keep their doors open.

I look forward to the flow of capital back into the market.  Until then, it’s going to be up to us as individuals to keep projects and businesses moving forward.  If you see a project or have a business you like, consider investing in those entities in addition to traditional investments.  I can assure you the gratitude from the business or project owner will far surpass that of a stock certificate, not to mention the potential returns may be very rewarding.
