On Thursday, Thanksgiving night, I was watching King 5 news. I was thinking I was only going to get updates about the shopping frenzy that was going to occur on Friday. Instead there was a small segment on social media fatigue. I had never heard the term before and tried to look for the news clip online, but was unable to find it. So I searched the internet and got a definition and use of the word. We are getting overwhelmed with social media and all of these networking sites. Even if it is a good way to keep in touch with friends and family, it is difficult to keep up with all of our accounts. I wonder if social media fatigue will cause networking sites to lose popularity.

Well, the wife did something yesterday that she hasn’t done much of in the previous few months… use me for sex.  It was very nice, to say the least.  Afterwards I mentioned to her that when I wished her a “happy anniversary” last week she had mentioned we that we needed to talk about our arrangement.  I could tell at the time it was nothing too serious.  It just sounded like a “we haven’t talked about it in a while” type of thing, but we have both been sooooo busy (for a couple of months now it feels) that yesterday was the first time I could bring it up.

Basically she ended up apologizing for not being more proactive.  I, of course, told her it’s not about her having to do things for me, but she is far smarter than that.  She understands that the Arrangement has to be a two way street.  As she said, “lately you’ve been doing all the cleaning and working around the house and not having sex… if we didn’t have the arrangement it would be the same thing”.  Her point wasn’t that the arrangement is pointless, but if she isn’t involving herself in doing things than it is no different than not having the arrangement.  She of course brought up giving me assignments again, which she hasn’t been doing.  I again explained that it wasn’t supposed to be a chore for her, but her point of view is that it personalizes my regular cleaning routine, plus the things she sees that she wants me to clean I may not see.  The problem is she doesn’t seem to have time to give me the assignments any more.  Not that I’m not cleaning, etc.  She acknowledged my cleaning.  Her point is about her involvement.

She seemed to get a little down at times during the conversation, but pointed out, without any prompting from me, that she didn’t want to take a break or end the Arrangement.  She did say she was going to make more of an effort to be involved, which is a wonderful thing, and the conversation ended on a good note.

To lighten the mood a bit at the end I gave her some orgasm statistics.  For me to have the same number of orgasms this year as I did last year I’ll need to have one every four days for the rest of the year.  She laughed and assured me that wasn’t happening.  She did throw me for a loop when she asked what her stats were for the year.  I was caught off guard and pointed out that it was easy to count my few orgasms to her many, but (I’m a dork) she knows that I track both.  I commented that she is probably off her pace from last year a bit as we’ve had a couple of slow months this fall.  However, when I went back and counted she is actually right on track with last year (of course that’s a rough estimate as I never know exactly how many times she cums).

All in all it’s been a very good week.  Anniversary, her birthday, Thanksgiving, using me for sex… good times!

SNPs genotyping analysis using Haploview

1. What is the name of haploview format to use in this analysis?

Answer: HapMap Project Data Dumps

2. Please show us the marker and individual quality control of the genotype data use in the analysis?

Answer: To analyze the SNP genotyping result from the question, we have to create the file which contain only “HapMap Project Data Dumps” information and then analyze it in Haploview software.
We can do so by copy the data provided in the question, but select only the SNP genotyping data. (Starting at “rs#” through to the end of last SNP marker)

Paste the data in a blank Notepad text file and saved it. (The file name shown here is “HapMap_Assignment.txt” ).

 At the Open new data menu, select “HapMap Format”. Then browse to the filed you saved. After that click OK.

The software will analyze the data and show the result in a windows with 4 tabs at the upper left corner. Select “Check Markers” tab, it will show the marker and individual control information. The marker datas are in the table, and the individual quality controls are at the bottom of table.

The marker’s details are:

• #  is the marker number.
• Name  is the marker ID specified (only if an info file is loaded).
• Position  is the marker position specified (only if an info file is loaded).
• ObsHET  is the marker’s observed heterozygosity.
• PredHET  is the marker’s predicted heterozygosity (i.e. 2*MAF*(1-MAF)).
• HWpval  is the Hardy-Weinberg equilibrium p value, which is the probability that its deviation from H-W equilibrium could be explained by chance.
• %Geno  is the percentage of non-missing genotypes for this marker.
• FamTrio  is the number of fully genotyped family trios for this marker (0 for datasets with unrelated individuals).
• MendErr  is the number of observed Mendelian inheritance errors (0 for datasets with unrelated individuals).
• MAF  is the minor allele frequency (using founders only) for this marker.
• Alleles  are the major and minor alleles for this marker.
• Rating  is checked if the marker passes all the tests and unchecked if it fails one or more tests.

The individual quality controls’ details are:

• Hardy-Weinberg p-value cutoff: >0.0010 (per population)
• Minimum genotype %: < 75% (per population)
• Maximum # Mendel errors: < 1 (per population; only applies to YRI, CEU, ASW, MEX, MKK)
• Minimum minor allele frequency: < 0.0010

Answer: The linkage disequilibrium (LD) map shows the strength of linkage disequilibrium between each SNPs. The more intensity of red color, the higher linkage disequilibrium value it is.

 This LD map obtained from the question has 30 markers. By using “Confidence Intervals” algorithm, there are 3 haplotype blocks in the map. The first block, 18 kb in size, consists of marker number 8 and 9. The second block, 80 kb in size, consists of marker number 13 – 17. Finally, the third block, 48 kb in size, consists of marker number 24-29. (See answer of the 4th question for the detailed on Haplotype block)

Answer: The haplotype display shows each haplotype in a block with its population frequency and connections from one block to the next. This represents the level of recombination between the two blocks (value of multiallelic D’). Above the haplotypes are marker numbers.

 To summarize, there are 3 haplotype blocks in the map.

• The first block consists of marker number 8 and 9.
• The second block consists of marker number 13,14,15,16 and 17.
• The third block consists of marker number 24, 25, 26, 27, 28 and 29.

Answer: A tagging SNP is a representative single nucleotide polymorphism (SNP) in a region of the genome with high linkage disequilibrium. It is possible to identify genetic variation by genotyping just only the tagging SNPs in the haplotype without genotyping every SNP.

To display tagging SNP, go to “Display” > select “Show tags in blocks”

Notice the grey triangles which will appear at the location of tagging SNPs in each haplotype block.

To summarize, tagging SNPs in each haplotypes are:

In class we have discussed which websites are best for viewing videos and we have talked about viewing television programs online. Often times creators make people pay to view episodes online or they don’t upload content online because they feel like they will lose ratings when people watch their episodes online. Therefore, people turn to “bootleg” sites where they get the content for illegally for free.

By posting their content online for free, the creators of South Park are ensuring that people don’t have to go anywhere else to watch their episodes. They have every episode available online and by doing this they are allowing viewers to watch more of the show and get interested enough to watch more episodes in the future. On Friday last week, I set about on my latest writing crusade. By the end of the week, I was happy with progress, and put down some aims for the coming week. By the end of the week(well almost) I have achieved two aims, but the ones I haven’t completed I am not too unhappy about.

Aims for this week:

• Work through next module and start on one after that for course work. [Done]
• Finish Chapter two, and get a good start on Chapter three of A Darkening Dawn [Not Done]
• Bonus to feel really virtuous – Finish Chapter three and start Chapter four. [Not Done]
• Word count aim: 3000 by Friday [Done]

I reached my word count aim, and discovered that chapter two is going to be much longer than chapter one. It is three and half thousand words and only half way there! Having not got this far into a story in ages it is hard to judge. But hay…

So aims for next week:

• Finish next module.
• Word count aim: 3000 words by Friday. Bringing total word count to 9000 words.

As for the EIC, she has been writing a children’s story aimed at our eldest boys age group. She is pleased because he liked it.

To factor the quadratic above:

1.)    Look at the constant of the quadratic, the last number, and lists its factors 1, 10; 2, 5; determine which set of factors when added give you the sum of the coefficient in the middle (2,5). These factors will be the second term in each binomial.

2.)    Look at the first term and factor it  x² = x ● x  these factors will be the first term in each binomial

3.)    Write the binomials (x + 2) (x + 5)

Writing the process does help with understanding factoring. I think that students would benefit from explaining their thinking once they have practiced the process several times with success. It would also be beneficial for the students to explain how they would check their work by working backwards form the binomial to the quadratic.

I have to say that Blogging is an interesting way to communicate with others. The thing I find difficult about it is that it takes time to go to and read peoples blogs. I had to Bookmark everyone’s from class so I could get to them easily, of course I had my favorite few that I would visit first before moving on. I am not a computer person. I check my e-mail at school daily, in the am and after lunch, I only check my personal e-mail every other day or so. As far as keeping this blog going I do not think I will take the time. I will keep an eye on my class mates to see if anyone enters new material and I may even leave a comment every now and then.

This class was interesting and I had to review some of the concepts covered. I have taught pre-algebra and can see that I may be asked to teach algebra I in the near future, since we have a 5th grader coming to the middle school for pre-algebra daily. It’s like that old saying, “it’s like riding a bike…” I have however shared many of the applets and several lessons with my math team. The function machine and the algebra quiz sites are two that I will definitely use in the classroom, because they will give the students a different perspective to the paper and pencil work.

I do have a math blog on my school web page. I have used it twice so far this year. The first blog the students told me what they like about math and did not like about math. My second blog is still posted because my daughter the marketing major needed my help with her senior project. I got permission to give the parents an internet survey that describes their child’s influence on them during back to school shopping. To complete the extra credit the students have to answer three questions about why the survey could be important; which average would give the best information, the mean, median, or mode; which graphs would they use to show the results and why. I told the students I would help them with a little extra credit since they are having their parents help my daughter. My daughter will then come in to answer these questions and more about math in her soon to be profession,  after the winter break. I do have some other questions to post later, but now I’m thinking I have some new material that may give more information about what my students know and don’t quite get. I am also using more writing in math this year, having students explain processes in words. I do not think I will use “journals” at least not separate from their notebooks. I may start having students write to prompts and have them turn in one each marking period that made an impression on them or when they looked back felt they made more progress with; not as a graded work but as a class participation grade.

I finally got my sleep after staying awake for more than 36 hours doing assignment! Yes, it’s last minute group work, but to be honest I think we did pretty well. One assignment down, 4 more to go!!

By the way, the protest in London is today and we (Ryan and I) are excited about it! We’ll be leaving for London around 9am, hopefully we’ll get some good shots! We’ll be reporting it right after the protest is done, so stay tuned!

The Sharehouse is a non profit agency that provides furniture and household goods free of charge to families that are in transition from homelessness too permanent housing. They collect furniture or household goods that you no longer want and deliver them to families that are in need of a couch, bed or table.

The Seattle/King County Coalition on Homelessness (SKCCH) provides an option for people or organizations to sign up as a member and they charge a set amount as a membership fee. These fees are used to “support the Coalition’s local, state, and national advocacy work, and help to maintain its 29 year history of organizing an independent, accurate, and community-driven One Night Count of people who are homeless and without shelter in King County” (SKCCH). Founded in 1979 they are one of the oldest homeless coalitions in the country.

Both of these links will be useful reference material for my project on Homeless Holidays in Seattle.

We were asked to complete a Semiotics Assignment based on a celebrity of our chosing using ideology. I choose Gay Byrne as my celebrity because I don’t really like him, he has always been well over-rated. I decided to portray him under the ‘I am Masculine’, ‘I am Badass’ and ‘I am Wealthy’. I choose photoshop as my medium of portraying this images.

I am Masculine

I wanted to portray Gay Byrne as masculine as possible so I decided to use wrestler ‘Kurt Angle’ as a basis for his body in this one. I received the Xbox 360 game TNA Impact which is a wrestling game and Kurt Angle is the centre focal point of the image. I cut around the head of Gay Byrne in my original image using the magnetic lasso tool and pasted his head upon Kurt Angle’s body. This made Gay Byrne phsyically look very masculine with a large wrestler’s body. Both wrestling and video games are a popular male interest so I thought this would be a good basis for Gay Byrne. I decided to take this further by placing a packet of comdoms on his right hand and a Gilette Razor on his left hand! I added 3 logos below the TNA Impact name, ‘Rated M for Messy’, ‘Spike TV Get More Action’ and EA Sports. The idea of being messy, being into action and sports in general is often conceived as being male. To finish off the image I cast a blue tint in the background around Gay Byrne which caused the 2 wrestlers either side of Gay Byrne to become alot less dominant as they are not important leaving Gay Byrne as the chief central  design element within the frame.

I am Badass/Evil
For this image I wanted to portray a setantic/hell environment. I imported the picture of Gay Byrne and changed his shirt to a black Tshirt and gave his T-Shirt a setanic 5-pointed star and a death metal band logo from ‘Emperor’. I also turned his face almost white creating more of a goth appearance to Gay Byrne. I blackened his lips and put red highlights into his eyes with a dark shadow under his eyes to futher capture the mood. I placed large horns on his head created by cutting them out of a background using magnetic lasso tool and placing them on his head. I painted some black near the base of the horns to give them a more realistic placing.

To create an appropriate background I got some hell fire and then placed another image on top of it of a demon and i blended the layers together using linear light blend modes.

I am Wealthy
For this I wanted to show Gay Byrne as rich as possible. I found a 100 euro note and decided to patch it making a pattern over the entire surface area of his shirt. I blended the notes into his shirt using the Difference blend modes and remoted the unwanted parts of notes outside his shirt by inverting selection and deleting the remaining notes. The notes blended nicely into his shirt. I choose the 100 euro note because it is green and I normally associate the colour green with money. I found the moneybag with dollers and placed it beside him and then I thought it would be a good idea to have a gold money safe beside the money and Gay Byrne and attached a gold credit card on the safe to further emphasis the aspect of having lots of money and being rich.

The elements of a euro notes wearing Gay Byrne, the moneybag and the gold safe were very strong and decided to have a less crowded image to fill the remainder of the image. I found a background doller sign which is patterned with small doller bills and placed this in the background and gave it a blue tint to create a contrast between it and the main focus of the image which is Gay Byrne, his money and the gold safe. This turned the image more asymmetrical.

Since my last post on project updates I have done a 360 turn on my project topic. Instead of doing plug-ins for WordPress I am now going to create a short video on the issue of “Homeless in Seattle.” I plan to create a short video in which I go around the city and try to capture the reality of living on the streets. I will link to sites like the Seattle’s Union Gospel mission in which people can donate to the cause and help provide food and shelter to these people. My time line is as follows:

Today is a holiday… and I still can’t go anywhere. There are two assignments waiting for me which haven’t been finished yet. And it’s so hard. One is a database design and another one is making a report about plagiarism. I’m completely confused, dunno where to start. For all these days, I only make a planning ’till now. But hopefully I can finish it soon as the due date is next Monday for database design and Wednesday for plagiarism report.

I wanna sleep… -_- I’m so sleepy…

Wow, I can’t believe now Friendster got a blog too!

Anyway, watta rainy day… Today I woke up with a headache.. I don’t know, was it becoz I always went back home in raining at last few days or was it becoz I was too tired finishing my assignments that made me had to sleep every 2 o’clock in the morning. Oh well…

But I’m glad now I’m free. Not really free actually, since there’s 2 new assignments and homeworks waiting for me but heyy… I need a break..  at least a couple of days.. Oh..hopefully I’ll get well soon, coz I can’t wait to go shopping tomorrow. Wanna join with me?

Today I finished my semiotics assignment. We were giving the task of creating three images using one celebrityI, each to show a certain statement from the list below

• I am masculine
• I am feminine
• I am wealthy
• I am popular
• I am evil
• I am Intelligent

I chose to create an image for I am Evil, I am Intelligent and I am wealthy. I chose Ross Kemp as my celebrity as I found an image of him that I thought could be easily edited to show my three choices.

The first image I created was the I am Wealthy image. The first thing that came to mind when i thought of wealth was money, I found an image of a ten pound note and then replaced the queens face with Ross Kemps face. I did this using Adobe PhotoShop CS4. I started by covering the queens facial features  using the clone tool and then overlaying a picture of Ross’ head over it. This kept the pattern in the money, making the photo look some what legit.

The next image I created was for the I am Evil statement. I decided to put Ross’ head onto Dr. Evil’s body from Austin powers as he is a well known character in pop culture. I then added devil horns onto his head  and a devil pitch fork to highlight the evil theme. I finally places this image over a picture of a burning house, implying that he burnt the house down.

The final image I made was for the statement I am Intelligent. I found an image of einstein writing equations on a board and thought it would look good if i put ross kemp in instead. I used the clone tool to remove the equations on the board so I could add my own. I also used the clone tool to remove most of Einstein’s head/hair and to add some shadow onto Ross’ head. I then overlayed text to look like chalk and added my own equations, I added E=MC2 as it is known by almost everyone and is directly associated with intelligence.
