Clairju: Introducing the "Melanin Diet" Theory

How does it work?

Most skin whitening agents target the inhibition of melanin synthesis through the enzyme tyrosinase. For this reason, these whitening agents are often called “tyrosinase inhibitors.” But in this novel “melanin diet” theory, it argues that melanin is synthesized because the skin needs it for protection against the adverse effects of sunlight.

On the other hand, the mechanism of action of Clairju targets the phagocytotic action of keratinocytes. Keratinocytes have phagocytotic action to eat synthesized melanin from the neighboring melanocytes. Because of this action, keratinocytes themselves turn dark (black).

Clairju works to “curb” the phagocytotic appetite of keratinocytes to devour synthesized melanin which are contained in melanosomes. Technically, this means the inhibition of uptake of melanosomes into the keratinocytes. Clairju can work in conjunction with other whitening agents or tyrosinase inhibitors to produce a highly effective whitening product.

Clairju is an extract obtained by hydrolysis of the fruit of Prunus cerasifera (Prunus domestica) Lindel. (Rosaceae) by water and used as skin care ingredient. Clairju is named as such with this in mind: “Fruit (extract) makes skin clear” by using clearness in French which is “clair” and jus which means fruit.

TECHNICAL INFORMATION

The prune fruits contain compounds such as beta-carotene, Beta-cryptoxanthin, lutein, neochlogenic acid Vitamins and minerals such as Vit. A, C, Eand B group.

Chime in: Back in the late ’90s when researchers and developers of whitening agents scrambled to come up with an effective skin whitening agent, many studies and possibilities of inhibiting the stages of the synthesis of melanin were made and looked into. One possibility then that they were looking into was the inhibition of the uptake of melanosomes into the keratinocytes. Back in those days, this was not yet a reality. But with the development of Clairju, that vision is now realized.

Whitening Theory

1.UV Absorption
When the mealanocye is actived by UV, inflammation etc, pigmentation is caused. UV not only stimulates directly melanocyte but also stimulates melanocyte through active oxygen, DNA damage, inflammation substances etc.

2.Inhibition of IL-1α Activity
When the skin is exposed to UV, IL-1α a cytokine which induces inflammation, is released from the keratinocytes in the epidermis, and IL-1α then stimulates keratinocyte endothelin release.

3.Inhibition of Tyrosinase Activity
In the melanocytes, tyrosine is metabolized (by mediation of tyrosinase) and melanin is generated.

4.Inhibition of Autoxydation-polymerization of Melanin and Reductive De-colorizing Action
Of the above-reactions, when DOPAquinone changes Eu-melanin via Leuco DOPAchrome, they react by autoxydation; black colored melanin is produced at first.

5. Anti-Oxidation
After making melanin, melanin polymer becomes larger because each melanin is joined joint by oxidation.

6.Repairing of DNA damage by UV
When people get UV, damaged DNA might be produced. Then, if damaged DNA is not repaired, sometimes abnormalities are produced. So, it is important to repair DNA damage by whitening.

7.Promoting of Producing of Pheo-melanin
Two type of melanin are known. One is Eu-melanin and the other is Phe-melanin. The color of the former is black and latter is yellow. So, when Pheo-malanin is produced instead of Eu-melanin, the skin looks white compared with black.1)

8.Inhibition of Producing melanin
When N-Acetyl-L-Tyrosine reacts with Tyrosine, an unknown yellow substance is produced instead of melanin.

9.Cell Proliferation When people get old, the speed of turn over is slower. The cell proliferation depression is one of the reasons.
